چکیده

Immunogenic cell death (ICD) is accompanied by changes in the composition of surface and release soluble mediators. Such signals operate on dendritic cells to stimulate presentation tumor antigens T cells. We recently found that low expression glucose-6-phosphate dehydrogenase (G6PD), a cytoplasmic enzyme involved pentose phosphate pathway, correlates with active anti-tumor immunity. G6PD maintains NADPH levels protects from oxidative damage. The present study demonstrates inhibition induces ICD as mechanism for activating immunity, intending develop combination treatment malignant melanoma. Chemical or genetic increased hydrogen peroxide-induced releasing high mobility group box 1 (HMGB1). Immunocytochemistry revealed calreticulin migrated dying Comprehensive gene analysis 83 melanoma samples NCBI GEO database showed causes upregulation endoplasmic reticulum stress-related genes. These findings indicate immunogenic death. next investigated efficacy therapy involving blockade anti-PD-1 antibody administration using mouse model. C57BL/6 mice transplanted G6PD-knockout B16 treated significant reduction size. In clinical settings, among 42 patients immune checkpoint inhibitors, those had better prognosis than (p=0.0473). not only promising biomarker but also new therapeutic target. Inhibition tumors death, which increases antigen presentation, thereby increasing effects inhibitors.

INHIBITION OF HUMAN ERYTHROCYTE GLUCOSE 6-PHOSPHATE DEHYDROGENASE ACTIVITY BY DEHYDROEPIANDROSTERONE AND RELATED STEROIDS.

The inhibitory effects of several steroids on G6PD activity using intact erythrocytes are reported. Incubation of whole blood with dehydroepiandrosterone (DHEA) resulted in 42% and 12% inhibition in the enzyme activity in the presence and absence of oxygen, respectively. Addition of epinephrine and/or aminophylline into the incubation medium caused further enzyme inhibition suggesting a po...

Importance of glucose-6-phosphate dehydrogenase activity in cell death.

The intracellular redox potential plays an important role in cell survival. The principal intracellular reductant NADPH is mainly produced by the pentose phosphate pathway by glucose-6-phosphate dehydrogenase (G6PDH), the rate-limiting enzyme, and by 6-phosphogluconate dehydrogenase. Considering the importance of NADPH, we hypothesized that G6PDH plays a critical role in cell death. Our results...

The Efficacy of Vitamin E and Folic acid on the Acute Hemolysis Caused by Glucose-6 phosphate Dehydrogenase

Background: Glucose-6 phosphate dehydrogenase (G6PD) deficiency is the most common inherited enzyme deficiency of red blood cells involving the enzyme pathway of hexose monophosphate. This study was conducted to examine the effect of vitamin E and folic acid on the improvement of acute hemolysis caused by the G6PD deficiency in patients referred to 17 Shahrivar Hospital, Rasht. Materials and M...

Inhibition of glucose 6-phosphate dehydrogenase by adenosine 5'-triphosphate.

D-glucose 6-phosphate dehydrogenase (G6PDH) catalyzes the oxidation of glucose 6-phosphate (G6P) to 6-phosphogluconate-6-lactone and thereby constitutes the step which introduces hexoses into the "pentose cycle" pathway. This well-known enzyme, which is widely distributed in nature, was purified to varying degrees from different sources (for literature see refs. 1 and 2). The yeast enzyme,3 4 w...
